Test item: Determination of bacterial growth curve

Bacterial growth curve refers to single-celled microorganisms. It is to inoculate a small amount of single-celled microorganisms into a certain volume of liquid medium, culture them under suitable conditions, and regularly sample and measure the number of cells. Take the logarithm of the number of cells growing as the vertical coordinate and the culture time as the horizontal coordinate to draw a curve. We call this curve the bacterial growth curve.

Determination steps

(1) Bacterial liquid culture: Pick a ring of fungus moss on the inclined surface culture of the strain, inoculate it in a thousand broth medium, and culture it at 3 TC for about 12 hours. This bacterial liquid is the seed culture liquid.

(2) Divide the medium and correct the zero point: Pipette 25 mL of medium with a sterile pipette and add it to a triangular flask with a side arm tube. Pour the uninoculated medium into its side arm tube and adjust the zero point on the photoelectric colorimeter. Even if the OD value on the photoelectric colorimeter is above zero.

(3) Inoculation and zero-hour determination: Pipette the 2. SmL seed culture solution into the inoculation bottle and shake it well. Pour the freshly inoculated culture solution into the side arm test tube and measure the OD value of the photoelectric colorimeter. The reading at this time is the zero-hour reading value in the growth curve of the strain after inoculation (ie, inoculation wipe).

(4) Cultivation and growth measurement The triangular flask measured at 0 o'clock is immediately placed on a constant temperature water bath shaker for culture. The culture temperature is 3 TC, and the frequency of the shaker is about 100 times/min (the temperature and frequency can be adjusted at any time). During the culture, the triangular flask should be removed from the shaker every 30 minutes, the bacterial liquid should be poured into the side arm test tube, and the OD value should be read on the photoelectric colorimeter. Record the measured data each time. During each measurement, use the culture solution of a blank control tube to correct the zero point of the photoelectric colorimeter.

(5) Drawing the growth curve: taking the measured time as the abscissa, the logarithm of the number of bacteria (OD value) as the ordinate, and drawing points on the semi-logarithmic coordinate paper, the resulting curve is the growth curve of the measured shade under experimental conditions.
