test flow
1. Appropriately dilute the HFMD virus suspension and inoculate it into Vero cells.
2. Add an appropriate amount of cell maintenance medium, place it in a 37 ° C, 5% carbon dioxide cell incubator, and observe CPE daily, usually for 3 to 5 days.
3. When it is found that the cells start to fall off in large groups from the bottom of the culture bottle, and 75% of the cells appear CPE, immediately freeze and thaw the cells three times (alternately at -20 ° C and 37 ° C), suck the virus suspension into the centrifuge tube, and centrifuge for 6000r/min for 10min to remove the precipitation.
4. After centrifugation, take the supernatant, put it in a cryopreservation tube, and store it in a refrigerator at -80 ° C.
5. Calculation of viral infection titer by endpoint dilution method.
6. Evaluation of disinfection effect: According to the use concentration (intensity) and action time specified in the product instruction manual, the test is repeated 3 times. The inactivation log value of each test is ≥ 4.00, which can determine that the inactivation effect of the product on hand, foot and mouth disease virus meets the disinfection qualification requirements; the positive control group virus titer log value should not be less than 6.00.
