This study uses a combination of flocculation and alcohol precipitation to refine Shuanglonglian, hoping to improve the efficacy and reduce the dose.

In order to improve the therapeutic effect of Shuanghuanglian extract, we first use supercritical extraction to extract volatile components. Using a uniform design method, the active ingredients of Shuanghuanglian can be better extracted by sub-boiling water extraction. Using orthogonal experiments, the purification of Shuanghuanglian is realized by a combination of flocculation and alcohol precipitation. In the final preparation process, volatile components are added to ensure its curative effect. In the flocculation treatment, the addition of flocculants can remove most of the protein, pectin, tannin and other colloidal components in the liquid through adsorption bridging and electric neutralization, while the polysaccharides and chlorogenic acid and other components are better retained. Through alcohol precipitation treatment, the active ingredients in the liquid such as chlorogenic acid are soluble in both alcohol and water, and the insoluble components in the liquid, such as polysaccharides and remaining chitosan flocculants, can be precipitated in the aqueous alcohol solution to remove the insoluble components in the liquid, such as polysaccharides and remaining chitosan flocculants, etc., to further purify the liquid, greatly reduce the product yield, and increase the content of the active ingredient chlorogenic acid. The obtained extract has been verified by further animal experiments, and good results have also been obtained, ensuring the curative effect and reducing the dosage, so that the improvement of the dosage form from oral liquid to capsule can be realized.
