3,3′,5,5′-四甲基联苯胺已用作竞争法和间接法酶联免疫吸附分析(ELISA)的底物。3,3′,5,5′-四甲基联苯胺(TMB)是一种二氨基联苯胺的非致癌性取代物(Ames测试阴性)适合在ELISA实验中作为过氧化物酶底物。底物产生可溶性终产物,淡蓝色,分光光度检测在370或620-650nm可以读峰值。TMB反应能被2M H2SO4终止(产生黄色),在450nm处可读。一种高敏感,特异性试剂用于检测血液,血红蛋白实验,过氧化物酶实验。 3,3′,5,5′-Tetramethylbenzidine has been used as a substrate in competitive enzyme-linked immunosorbent assay (ELISA) and indirect ELISA. 3,3′,5,5′-Tetramethylbenzidine (TMB) is a noncarcinogenic substitute (Ames test negative) for benzidine suitable as peroxidase substrate for use in ELISA procedures. The substrate produces a soluble end product that is pale blue in color and can be read spectrophotometrically at 370 or 620-650 nm. The TMB reaction may be stopped with 2 M H 2 SO 4 (resulting in a yellow color), and read at 450 nm. A sensitive and specific reagent for the detection of blood, assay of hemoglobin, assay of peroxidases.
