Cystathionine-β-lyase (CBL, EC 4.4.1.8) represents a pivotal member of the lyase family, demonstrating exceptional substrate specificity. The enzyme catalyzes the β-cleavage of L-cystathionine with high precision, generating homocysteine (Hcy) through the sequential elimination of pyruvate and ammonia moieties. Reaction Synergistic Catalytic System with CBS‌ 1.CBL forms an efficient catalytic tandem with cystathionine-β-synthase (CBS, EC 4.2.1.22, Aladdin No. C774059) in the cyclic Hcy detection mechanism: ‌2. Condensation Phase ‌: CBS initiates the reaction by condensing Hcy with serine to form cystathionine. 3.Cleavage Phase ‌: CBL then cleaves the cystathionine molecule to regenerate Hcy. This cyclic amplification strategy enables highly sensitive quantitative Hcy determination. Consequently, CBL serves as the cornerstone enzyme in cystathionine-based Hcy detection kit development while also finding utility in other enzymatic applications requiring β-specific cleavage catalysis.
