肌酸激酶(Creatine Kinase M, CKM)是一种参与能量稳态的细胞质酶,编码的蛋白质能可逆地催化 ATP 和各种磷酸原,如磷酸肌酸之间的磷酸转移。它在大脑和其他组织中充当同二聚体,在心脏中充当具有类似肌肉同工酶的异二聚体。在 RPA 技术中,该酶可用于构成 RPA反应能量供应体系,利用肌酸激酶催化生成的 ATP 水解供应能量,改变重组酶 (T4 Uvs X) 与引物形成的复合体构象,使重组酶解离暴露出引物的 3’ 端,促进链置换 Bsu DNA 聚合酶结合至引物上,提高 DNA 扩增效率。 用途 RPA 反应能量供应体系 用于骨骼肌疾病及心肌疾病的诊断 推荐使用条件 储存缓冲液:20 mmol/L Tris-HCl (pH8.0), 150 mmol/L NaCl, 0.5 mmol/L EDTA, 0.5 mmol/L DTT, 50%甘油。
