​ 抗荧光淬灭剂是一种能够减缓荧光淬灭的试剂,可用于大多数的荧光染料中,操作简便,该产品推荐运用于固定透化的细胞或者组织中,观察结束后可使用指甲油或者密封胶密封盖玻片边缘;含 DAPI 的抗荧光淬灭剂可直接进行细胞核染色。 使用方法: 注意:产品使用前,将其恢复至室温,轻微震荡混匀,避免产生气泡。 1. 细胞样品: (1)染色完毕后,吸掉液体。(2)每个玻片上滴加 10-20 μL 的抗淬灭剂,盖上贴有细胞的盖玻片,让细胞接触到抗淬灭剂,盖上盖玻片时可以挤出多余 的抗淬灭剂,尽量不要有气泡。(3)荧光显微镜观察。(4)(可选):观察结束后,可使用指甲油或者密封胶密封盖玻片边缘。 2. 组织切片: (1)染色完成后,吸掉染色液。(2)滴加 10-20 μL 的抗淬灭剂于组织切片上,盖上盖玻片,让切片接触抗淬灭剂,盖上盖玻片时挤出多余的抗淬灭剂,尽量避免气泡产生。(3)荧光显微镜观察组织切片。(4)(可选):观察结束后,可使用指甲油或者密封胶密封盖玻片边缘。 3. 其他样品:其他样品可参照上述样品进行操作。 注意事项: 1. 抗荧光淬灭剂对于染色活细胞膜染料和线粒体染料的抗淬灭效果不佳,建议抗荧光淬灭剂用于固定透化的细胞或者组织。 2. 抗荧光淬灭剂与我们的 YF 488 染料搭配时,有可能产生背景干扰,这种情况可能是加入过多本品造成,应尽量减少本品的加入量。 3. 抗荧光淬灭剂可能会不适用于某些染料,实验前建议进行预实验测试匹配性。 4. 为了您的安全和健康,请穿实验服并戴一次性手套操作。 应用范围: 抗荧光淬灭剂 Anti fluorescence quencher is a reagent that can slow down fluorescence quenching. It can be used in most fluorescent dyes with simple operation. This product is recommended to be used in fixed permeabilized cells or tissues. After observation, nail polish or sealant can be used to seal the edge of the coverslip; DAPI containing antifluorescent quencher can directly stain nuclei. Presentation: Before using the product, return it to room temperature, slightly shake and mix to avoid bubbles. 1.Cell samples : ( 1 ) After dyeing, absorb the liquid. ( 2 ) Add 10-20 μL anti-quenching agent to each glass slide, cover the cover glass with cells, and let the cells contact the anti-quenching agent. When the cover glass is covered, the excess can be squeezed out. The anti-quenching agent, try not to have bubbles. ( 3 ) Fluorescence microscope observation. ( 4 ) ( optional ) : After the observation, nail polish or sealant can be used to seal the edge of the cover glass. 2. Tissue sections : ( 1 ) After the staining is completed, the staining solution is sucked off. ( 2 ) Drop 10-20 μL of anti-quenching agent on the tissue section, cover the cover glass, let the section contact the anti-quenching agent, and squeeze out the excess anti-quenching agent when cover the cover glass to avoid bubbles as much as possible. ( 3 ) Observation of tissue sections by fluorescence microscope. ( 4 ) ( optional ) : After the observation, nail polish or sealant can be used to seal the edge of the cover glass. 3.Other samples : Other samples can be operated with reference to the above samples. Matters needing attention: 1. the anti fluorescence quencher has poor anti quenching effect on staining live cell membrane dyes and mitochondrial dyes. It is recommended that the anti fluorescence quencher be used to fix permeabilized cells or tissues. 2. when the anti fluorescence quencher is matched with our YF 488 dye, there may be background interference. This situation may be caused by adding too much of this product. The amount of this product should be minimized. 3. the anti fluorescence quencher may not be suitable for some dyes. Pre experiment is recommended to test the matching before the experiment. 4. for your safety and health, please wear experimental clothes and disposable gloves.
