UltraBio™ 是一款专为低温储存(-80℃/-196℃)各类哺乳动物组织样本和细胞设计的、化学成分明确、即用型冻存液，含有 10% DMSO。冻存复苏后，细胞活率高达90%以上，极敏感细胞活率亦可大于 75%。 • 无血清 • 无动物源成分 可冻存组织/细胞种类包括： • 全血样本 • 实体组织样本 • 类器官 • 多能细胞(iPSC、MSC、HSC 等） • 原代细胞（人/动物免疫细胞） • 癌细胞和常规细胞系（骨髓瘤细胞、杂交瘤细胞、CHO和 HEK293 等） 使用方法 1. 样本准备 [实体组织和类器官] a)（如需要）使用 OPM 组织清洗液 或 PBS 清洗组织样本/类器官。 b) 酶解类器官 / 将实体组织切成小于 1 mm 的小块*。 [待冻存组织样本体积越小，复苏后细胞活率越高]。 c) 按 1:500（样品：UltraBio™ v/v）加入预冷的冻存液（2-8℃）。 [全血样本] a) 使用肝素抗凝管采集全血样本。[不建议使用 EDTA 抗凝管采集] b) 按 1:19 （样品：UltraBio™ v/v）加入预冷的冻存液（2-8℃）。 [细胞样本] a) 离心收集细胞沉淀（800 rpm, 5 分钟），弃去上清。 b) 加入预冷的冻存液（2-8℃）重悬细胞，冻存细胞密度推荐 0.5-3 x 106 cells/ml。 2. 冻存 将样本分装到冻存管中，按如下操作进行冻存： • 2-8℃ 孵育 10-15 分钟. • 将冻存管放置于程序性降温盒（冷却速度 1℃/min）并放置在-80℃冰箱内。 • 如需长期保存，可转移至液氮罐中。 3. 复苏 • 取出冻存管。使用 70% 的乙醇或异丙醇擦拭外部。 • 在生物安全柜中将冷冻管盖旋转 1/4 圈，以释放内部压力，再重新拧紧。 • 在 37℃ 水浴中快速融化样品（ • 使用 70% 乙醇或异丙醇重新擦拭冻存管外部。按1:10 的比例（v/v）将解冻的样品转移至预热培养基中，轻轻混匀。 • 离心（800 rpm，5 分钟），弃上清。即可根据实验需求进行后续操作。 [实际离心速度和时间因细胞类型而异]。 注意事项 1. 请注意始终在无菌环境中进行操作。 2. 为了保护您的安全与健康，实验中请全程穿戴好实验服和一次性手套。
