阿拉丁的UltraBio™ Streptavidin Magnetic Beads (链霉亲和素磁珠),也称Streptavidin磁珠或SA磁珠,由高质量的链霉亲和素 (Streptavidin, SA)与超顺磁性纳米级磁珠共价偶联而成,能够快速、高效、灵敏、特异性地与生物素(Biotin)标记的抗体、核酸、蛋白、多肽、凝集素等分子结合。主要用于分离纯化生物素标记的核酸、抗体、蛋白或相关复合物等,用于免疫沉淀(Immunoprecipitation, IP)、细胞分选、DNA-蛋白相互作用研究等。 Streptavidin的分子量为 53.2kD,可以高度特异性地和生物素(Biotin)结合。Streptavidin和生物素的亲和常数为 Kd=10-15M。Streptavidin是一个4聚体蛋白,可以同时结合4个生物素分子。Streptavidin的中文名为链霉亲和素,从Streptomyces adidiniil中纯化获得,和鸡蛋清来源的Avidin(亲和素)在空间结构以及与生物素的亲和力方面具有高度的相似性。和Avidin不同的是,Streptavidin是一种非糖基化蛋白,并且基本不带电荷。Avidin的pI=~10.5,在中性pH条件下呈碱性。由于Streptavidin和Avidin相比在中性条件下不带电荷,因此Streptavidin的非特异性结合比Avidin低很多,这样检测时的非特异性背景就低很多。因此目前在生物素检测时通常使用Streptavidin替代Avidin。 链霉亲和素磁珠在生物医药领域内应用非常广泛,可以特异性地结合生物素标记的抗原或者抗体,作为免疫沉淀、细胞分选、ELISA等反应的载体;结合生物素标记的DNA或RNA片段,从细胞或组织提取物中分离特定的核酸-蛋白质复合物,用于蛋白质与核酸相互作用研究;结合生物素标记核酸探针,用于DNA、RNA杂交实验或mRNA的分离和纯化等;还可用于纯化单链生物素标记DNA寡核苷酸、分离生物素标记PCR产物等。 产品优势 结合容量高: 对复杂样品中生物素标记的分子可以快速进行分离纯化,而且磁珠粒径小,不易产生非特异吸附,每毫升磁珠悬浊液含约10mg磁珠。 特异性强: 可特异性地结合生物素化的抗体、核酸、蛋白、多肽、凝集素等配体分子,获得的产物纯度高,可进一步用于Western、ELISA、Northern、qPCR、质谱分析等一系列后续的分析测试。 结合生物标记分子速度快,吸附时间短,可快速高效结合配体。 所使用的纳米级磁珠(~200nm)具有超大比表面积,有效缩短了链霉亲和素与生物素结合所需的时间。缩短操作时间可以避免在长时间操作过程中配体分子的降解,有效保持配体分子的活性及完整性。 注意事项 1. 磁珠取用前应充分混匀,防止取用改变磁珠浓度,避免长时间超声对磁珠表面破坏; 2. 磁珠取出后,使用前请进行磁分离并用纯水或所用缓冲溶液清洗2-3遍; 3. 磁珠使用和保存过程中应避免冻融。 Aladdin's UltraBio™ Streptavidin Magnetic Beads are superparamagnetic beads, with 200nm particle size, covalently coupled with high-quality streptavidin that binds with biotin ligands specifically, rapidly, and highly efficient. The beads can be used for immunoprecipitation (IP), protein interaction studies, DNA-protein pulldowns, and purification of biotin-labeled proteins and nucleic acids. Streptavidin coupled to UltraBio™ Magnetic Beads has a mass of 55kD and 4 biotin-binding sites. It binds to biotin with extremely high affinity (Kd=10-15M). Streptavidin is a functional and structural analog to avidin. However, unlike avidin, streptavidin is non-charged due to the absence of carbohydrate groups and its near-neutral isoelectric point (PI), resulting in low nonspecific binding under neutral conditions. Therefore streptavidin instead of avidin is more regularly used in biotinylation-relevant studies. UltraBio™ Streptavidin Magnetic Beads bind with biotin-labeled proteins and nucleic acids, and have wide applications in biomedical studies, such as acting as a carrier for IP, cell sorting and ELISA, isolating DNA/RNA-protein complex, DNA/RNA hybridization, isolating mRNA, ssDNA oligonucleotides and PCR products, etc. Product Advantages Compared to most other similar products, the UltraBio™ Streptavidin Magnetic Beads bind with biotin-labeled molecules in complex biological samples with higher capacity and lower nonspecificity due to its more condensed streptavidin coupling and smaller particular size. Per milliliter of the beads suspension contains 10mg of magnetic beads. UltraBio™ Streptavidin Magnetic Beads bind with biotin-labeled molecules rapidly and high efficiently. The nanometer magnetic bead (~200nm) has a large specific surface area, facilitating a fast and efficient binding between streptavidin and biotin-labeled molecules. A shorter operation time can effectively minimize the degradation or denaturation of biotin-labeled molecules. Because of the low nonspecific binding of UltraBio™ Streptavidin Magnetic Beads, the highly purified biotin-labeled molecules are compatible with mass spectrometry analysis. Precautions 1. Before using magnetic beads, they should be thoroughly mixed to prevent changes in the concentration of the beads and to avoid prolonged ultrasonic damage to the surface of the beads; 2. Before using magnetic beads, please perform magnetic separation and clean 2-3 times with pure water or the buffer solution used; 3. During use and storage, avoid freezing and thawing.
