Product description: 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyltetrazolium Bromide (MTT) is a type of tetrazolium salt that changes to formazan when taken up into living cells. As formazan is a non-water soluble crystal, it can be dissolved in DMSO and the absorbance at 570 nm can be used to count the number of cells. This product consists of 5.0 mg / mL MTT dissolved in PBS. When 1/10th of the medium is added to cultured cells, the live cells are properly stained. Product Information: Volume/Quantity : 1mL x 5 Product Form : 5.0 mg / mL in PBS Directions for Use: MTT solution can be added at any point in the cell culture. For best results, it should be added to cells in the logarithmic growth phase. Pre-determine the number of cells and seed 10,000 to 100,000 cells per well in a 96-well plate. 1. Seed 100 μL of cells in a 96-well plate. 2. Incubate overnight in an incubator. 3. Add 10 μL of MTT solution to each well. 4. Incubate for 2-4 hours for color reaction. 5. The medium is removed, taking care not to inhale the formazan. 6. Add 100 μL of DMSO (Product No. D0798) and dissolve the formazan. 7. Measure the absorbance at 570 nm. Storage: This product should be stored at -20 °C and should be protected from light. Avoid repeated freezing and thawing.
