尿嘧啶-N-糖基化酶(UNG酶)也称为尿嘧啶DNA糖基化酶(UDG酶),通过大肠 杆菌表达纯化的重组酶,该蛋白分子量为25kD,可催化含尿嘧啶的单链和双链DNA释放 游离尿嘧啶,并且对RNA无活性,主要应用于防止PCR扩增产物的污染。该酶作用机理 为:在PCR反应中以dUTP代替dTTP,扩增产物片段中的T全部被U取代,形成了含dU 碱基的PCR扩增产物。UNG酶能选择性断裂单链和双链DNA中U碱基的糖苷键,降解反 应体系中含U的DNA,有效消除PCR产物的残留污染,大大降低扩增产物污染导致的假 阳性,从而保证扩增的特异性和准确性。 组分
