氨曲南专用酶是采用重组基因的水溶性蛋白,呈深褐色液体。根据氨曲南的化学结构而构建的重组基因蛋白。其基因片断的活性中心,可破坏氨曲南的化学结构式,使开环/断链后失去该药物的 抗菌性。 做抗生素的无菌检查时,应用手工注射器吸取无菌水注入药物的西林瓶中,摇匀,溶解,再吸出。加入至0.9%氯化钠的500ml溶液中,摇匀,不可用培养器上的针头溶解,输入,避免高浓度的溶液 通过滤膜,造成难以冲洗彻底。 这里,再次强调:做抗生素的无菌检查时,一定要手工注射器 注入无菌水溶解样品,再把溶解的样品转移至0.9%氯化钠的500ml溶液中摇匀,使得样品不会造成局部浓度过高,通过滤膜时难以冲洗彻底的情况。 无菌检验时,取酶2ml加入无菌水3ml,摇匀,使成氨曲南 专用酶的稀释液,取2ml酶的稀释液放入1500ml的冲洗液摇匀。待冲洗液冲洗培养器的滤膜完成后,泵尽。用手工注射器在三个培养器的呼吸口刺入,各加1ml的稀释酶,尽量让酶平铺在滤膜整个面,静止10分钟,使高浓度的酶与培养器滤膜充分接触,以破坏(中和、灭活)滤膜上残留的氨曲南药物,然后再泵入相应的培养基,摇匀。阳性对照管加入1ml的对应试验菌。 在冲洗液加2ml的氨曲南专用酶的稀释液,可以去除滤膜内部 残留的氨曲南的少量药物抗菌性。 在三个培养器加1ml的氨曲南专用酶的稀释液,可以去除培养器内部壁上和滤膜表面残留氨曲南的少量药物抗菌性。 每天上午、下午各对阳性对照管轻轻摇匀一次。 客户可根据上述做方法学验证,也可根据实际操作情况做验证。 The special enzyme of amtronam is a water-soluble protein using recombinant gene, which is dark brown liquid.
