本公司生产的 T4 多聚核苷酸激酶(3'磷酸酶活性缺失),即 T4 Polynucleotide Kinase (3' phosphatase minus),简称 T4 PNK (3' phosphatase minus)是一种多聚核苷酸 5'羟基激酶,可以催化 ATP 的γ位磷酸基团向单链或双链 DNA、RNA、寡核苷酸或带有 3'磷酸基团的单核苷酸的 5'羟基转移。其它 NTP 也可产生相同的反应:5'-OH + NTP → 5'-P + NDP。与 T4 Polynucleotide Kinase 相比较,T4 Polynucleotide Kinase (3' phosphataseminus)缺失了 3'磷酸酶活性,即不能催化 3'端磷酸化多聚核苷酸的去磷酸化:3'-P → 3'-OH + Pi (最适 pH 为 5.9 左右)。T4 PNK (3' phosphataseminus)特别适用于把 OH-DNA/RNA-p 催化转变为 p-DNA/RNA-p,或把 Np 催化转变为 pNp。 来源(Source)
