DNMDP, a phosphodiesterase 3A (PDE3A) inhibitor, is a potent and selective cancer cell cytotoxic agent. DNMDP binding to PDE3A promotes an interaction between PDE3A and Schlafen 12 (SLFN12). DNMDP shows clear cell-selective cytotoxicity In Vitro DNMDP (0-1000 μM; 48 hours) shows clear cell-selective cytotoxicity. HeLa cells undergo apoptosis upon DNMDP treatment, indicates by caspase activity detected by a caspase-sensitive luciferase assay and by poly ADP ribose polymerase (PARP) cleavage. At a concentration of 100 nM, DNMDP specifically inhibits both PDE3A and PDE3B, weakly inhibits PDE10, and had little or no detectable effect on other phosphodiesterases. MCE has not independently confirmed the accuracy of these methods. They are for reference only. Cell Viability AssayCell Line: NCI-H1563, NCI-H2122, HeLa, A549, MCF7, and PC3 cells Concentration: 0-1000 μM Incubation Time: 48 hours Result: Showed an EC 50 between 10 and 100 nM for two additional lung adenocarcinoma cell lines, NCI-H1563 and NCI-H2122, and for HeLa cervical carcinoma cells, but an EC 50 greater than 1 µM for A549, MCF7, and PC3 cells.
