本公司生产的核酸内切酶 VIII,即 Endonuclease VIII 来自大肠杆菌,是由阿拉丁自主研发的技术平台表达、纯化获得的重组内切酶。Endonuclease VIII 是一种 DNA 损伤修复酶,具有 N-糖基化酶(N-glycosylase)活性和 AP 裂解酶(AP-lyase)活性。Endonuclease VIII 的 N-糖基化酶活性能切下并释放双链 DNA 上受损的嘧啶碱基(Pyrimidines)从而产生一个脱嘌呤(Apurinic, AP)位点;Endonuclease VIII 的 AP 裂解酶活性,可以切割 AP位点的 3'和 5'端,从而切除 AP 位点,产生一个具有 3'和 5'磷酸的碱基缺口(Gap)。可以被Endonuclease VIII 识别并切除的受损碱基包括:Urea、5,6-dihydroxythymine (5,6-二羟基胸腺嘧啶)、Thymine glycol (胸腺嘧啶乙二醇)、5-hydroxy-5-methylhydantoin (5-羟基-5-甲内酰脲)、Uracilglycol (尿嘧啶乙二醇)、6-hydroxy-5,6-dihydrothymine (6-羟基-5,6-二氢胸腺嘧啶)和 Methyltartronylurea (甲基羟丙二酰脲) 。虽然Endonuclease VIII 与 Endonuclease III 相似,但 Endonuclease VIII 具有β和δ裂解酶活性,而 Endonuclease III 仅具有β裂解酶活性。 组分和说明 E745665
