本公司生产的T4RNA连接酶2截短型(T4Rnl2,truncated或Rnl2(1-249)),该酶仅包含T4RNA连接酶2N端249个氨基酸,可特异性地将5'端预腺苷酰化的DNA或RNA(App-DNA或App-RNA)连接到RNA的3'羟基。T4RNALigase2,truncated与T4RNALigase2的酶活性完全不同,后者是一种ATP依赖的dsRNAligase,而前者是一种不依赖于ATP的,依赖于5'端预腺苷酰化的单链核酸连接酶。T4RNALigase2,truncated常用于microRNA(miRNA)等3'端为羟基的单链RNA在克隆、高通量测序建库或PCR检测等时,在3'羟基端添加5'端预腺苷酰化的DNA或RNA接头。T4RNALigase2,truncated只能利用5'端预腺苷酰化的单链DNA或RNA作为3'端接头,因此可以大大降低连接反应的背景,通常是小RNA等建库时的优先选择。T4RNALigase2,truncated连接时不需要ATP,但需要5'端预腺苷酰化(也称预腺苷化、腺苷酰化或腺苷化)底物。即使在有ATP存在的情况下,T4RNALigase2,truncated也不能催化连接单链DNA或RNA5'-P末端与3'-OH末端之间形成磷酸二酯键。而T4RNALigase1可以在ATP存在的情况下,催化连接单链DNA或RNA5'-P末端与3'-OH末端之间形成磷酸二酯键。因此T4RNALigase2,truncated特别适用于smallRNAlibrary的制备,不管是进行miRNA的测序还是进行directionalmRNA-Seq,该酶都可以提高建库的质量,并有效降低连接反应的背景。 纯度(Purity):无 DNA 内切酶和外切酶活性,无 RNA 酶活性,无蛋白酶活性。 组分和说明 T750874
