蛋白内切酶Lys-C

赖氨酰内肽酶是一种水解非末端氨基酸的赖氨酰肽键的酶。它是高度特异性的，并且优先裂解Lys-I-Xaa键，包括Lys-I-Pro。在研究中用作酶消化的标准实验室试剂。这对于研究蛋白质的一级结构很有用。 本品为大肠杆菌重组表达赖氨酰内切酶，与天然无色杆菌蛋白酶有相同的酶活性。本品为重组表达产物，不含有其他杂蛋白酶（如 Lys-N、Arg-C等丝氨酸类蛋白酶），特异性针对赖氨酰肽键酶切，包括含DK、PK序列的完全特异酶切。 使用方法： （1）Lys-C与目的蛋白质量比=1:1000-1:20000，适宜pH为7.0-9.5，反应温度30-37℃，反应时间2-24小时。 （2）用户可根据研究目的，优化最适温度，反应时间和酶的比例。 （3）酶切体系中应避免 TLCK、盐酸胍等酶抑制剂。 （4）反应体系中含0.2%SDS、1.0%Triton、6mol/L尿素、0.5mol/L NaCl、20%乙腈对酶切反应均无影响。 产品特性： （1）产品性状：本品为白色或类白色冻干粉末，装量为0.5mg，1.0mg/支（按酶蛋白含量计）。 （2）酶含量：标示量的±10%（Lowry法，参照2015版中国药典）。 （3）电泳鉴定（SDS-PAGE）：为单一主条带，纯度不低于90%。 （4）电泳鉴定相对分子量为31±3.1 kDa。 （5）RP-HPLC纯度鉴定：面积归一法主峰纯度不低于90%。 （6）酶比活性：不低于3.0AU/mg。 Lysyl endopeptidase is an enzyme that hydrolyzes lysyl peptide bonds of nonterminal amino acids. It is highly specific and preferentially cleaves Lys-I-Xaa bonds, including Lys-I-Pro. Used in research as a standard laboratory reagent for enzyme digestion.
