利用绵羊细胞与相应抗体（溶血素）结合成的复合物，可激活血清中的补体，导致红细胞溶解，当红细胞和溶血素量一定时，在规定反应的时间内，溶血程度与补体量及活性呈正相关。在接近 50%溶血（ CH50）时，二者之间近似直线关系，故以 50%溶血作为最敏感的判断终点。以引起 50%溶血所需的最小补体量为一个 CH50U，可计算出待测血清中总的补体溶血活性，以CH50U/ml 表示。本实验主要反映补体经典活化途径的溶血功能，其结果与补体 C1～C9 各组成分的量及活性均有关。 试剂及配制 1、缓冲生理盐水： （1） 贮存液： Na2HPO4.12H2O2.85g KH2PO4 0.27g NaCl 17.00g 蒸馏水定容至 100mL，4℃保存备用。 （2）应用液：5mL贮存液加 95ml 蒸馏水，再加 10%硫酸镁 0.1mL，当日配制，12小时内使用。 2、2%绵羊红细胞：无菌采取绵羊血液，于等量 Alsever 液中可保存 3 周。用前以缓冲液洗 3次，并配成 2%细胞悬液。必要时可用吸光光度法或细胞计数法使悬液细胞浓度标准化。 3、溶血素：将绵羊红细胞溶血素（效价 1:4000），用应用液做 1:200 倍稀释（即 1mL溶血素加 199ml 应用液）。 4、待测血清：新鲜血液室温放置 30min，再 4℃放置 60min，使血块收缩， 4℃离心（ 3000r/min）10min，取上清，-20℃保存。 操作步骤 试管法（改良 Mayer 法） 1、致敏羊红细胞： 2%绵羊红细胞加等量稀释后的溶血素（1:2000），混匀，置 37℃水浴 30min。 2、稀释血清：待测血清 0.2mL，加应用液 3.8ml，稀释度为 1:20。 3、配制溶血标准管：全溶血管:2%绵羊红细胞 2mL加 8mL蒸馏水，混匀。50%溶血管:取全溶血管液 2mL加缓冲液 2mL。 4、按表所示，依次加各成分于试管中，混匀，置 37℃水浴 30min， 第 10 管为非溶血对照： 补体 CH50 测定试管法 成分
