3,3',5,5'-Tetramethylbenzidine (TMB)是一种过氧化物酶底物,用于ELISA 反应。底物产生一种可溶性淡蓝色的末端产物,可在370 或 655 nm在分光光度计上读取。TMB反应可以用2 M H2SO4 (产生黄色)终止,450 nm读取。 使用方法 使用之前,以超纯水反复洗涤的适当容器,倒出适量单组分TMB底物溶液,待溶液达到室温即可使用。 加液:加完HRP结合物并温育一定时间后,洗板3-5次,每孔加TMB底物100-150uL, 37°C 避光温育15-30分钟(可根据实验需求延长以获得预期显色)。 终止(备选):加等体积的1N的HCl或者2N的H2SO4来终止反应。酶标孔中反应液从蓝色变为黄色。 读数:终止反应后,30分钟内在450nm处读数。不加终止液,可以在620-650nm处读数。 注意: 降低底物强度:如果出现高的反应背景和/或沉淀,表明TMB底物反应过于强烈。为了避免产生沉淀,可以在加入终止液后马上读数;另外,可以进一步稀释一抗和/或HRP结合物。为了降低非特异性背景颜色,推荐进一步降低一抗和/或HRP结合物的浓度,尽量避免降低底物浓度。 动力学检测:TMB单组分底物在与过氧化物酶反应时产生一种蓝色,可以在加底物后620-650nm处读数,不需终止反应。 稳定性:2-8°C条件下,大于18个月。 敏感性:相当于显色AB双液,或 TMB因其灵敏度较高及无致癌性而得到广泛应用。单组分TMB过氧化物酶底物与辣根过氧化物酶连接物反应时产生一种深蓝色的产物,适用于定量以及定性分析的酶联免疫分析当中。 TMB is a substrate for horseradish peroxidase. Forms a soluble blue end product that may be read at 370 nm or 655 nm. The TMB reaction may be stopped with 0.5 M H2SO4, resulting in a yellow color, and read at 450 nm. Recommended for ELISA (microwell) procedures, not recommended for membrane applications. 3,3′,5,5′-Tetramethylbenzidine has been used in a study to demonstrate that by measuring substrate absorption values off the absorption maximum, the measuring range of any immunoenzymometric assay can be extended significantly without jeopardizing assay characteristics. 3,3′,5,5′-Tetramethylbenzidine has also been used in a study to understand the formation of CuS concave superstructures with peroxidase-like activity.
