由胃粘膜主细胞分泌的胃蛋白酶原，在PH 1.5～5.0条件下，经活化提取而成。为白色或淡黄色粉末。分子量约34500。易溶于水呈不同程度的乳白色，几乎不溶于醇、氯仿和醚。水溶液加热至70℃或在碱性溶液中其活性很快破坏，但其干燥品可耐热至100℃，具吸湿性，微酸及盐味。本品能使胃酸作用后凝固的蛋白质分解成胨，但不能进一步使之分解成氨基酸。其消化力以含0.2%～0.4%盐酸(PH=1.6～1.8)是为最强。
