AG 99是一种强效的和竞争性的EGFR酪氨酸激酶抑制剂,与底物polyGAT和ATP竞争[1].在血清饥饿的人膀胱癌细胞系5637细胞中,c-Met/肝细胞生长因子(HGF)受体的β亚单位,145-kDa蛋白,在1003\1234和1235酪氨酸残基上被磷酸化.此外,酪氨酸激酶EGFR和Src被激活.同时,AG99或Src抑制剂(PP2,SU6656)显著抑制p145met的酪氨酸磷酸化,并通过caspase样蛋白酶的活化诱导细胞死亡.这些结果表明,由Src和EGFR介导的p145met的酪氨酸磷酸化参与5637细胞的血清非依赖性生长。
