In the labeling process, simply mix the antibody and dye into the reaction buffer solution provided in the kit and incubate for a short time. Aladdin ® The dye no longer reacts at the end of labeling, so no further purification step is required. After labeling, about 4-6 molecules of dye are covalently bound to 1 molecule of antibody. Aladdin ® It is covalently labeled, so the antibody labeled with it can be used for multicolor fluorescent staining without dye transfer between antibodies. Aladdin ® The labeling can be compatible with NaN3, low concentrations of glycerol, Tris, and glycine. The micro ultrafiltration centrifuge tube provided in the kit can quickly remove incompatible small molecule antibody stabilizers before labeling. Standard Aladdin ® The labeling step can be used for more than IgG（ μ g) 4 times the BSA or gelatin samples. Select the kit size according to the amount of IgG YOu want to label. The optimized labeling step is suitable for IgG samples that stabilize excess protein or ascites. In this case, the kit size is selected according to the total amount of protein (IgG + stabilizer, or the total amount of protein in ascites) needed to label the antibody sample. The optimization step can also be used to label samples with IgG amount lower than the lowest range, and the total protein amount can reach the labeling range of the kit by adding stabilizer protein. The optimization step is not recommended for labeling natural antisera or hybridoma cell line culture supernatants because the antibody content of these samples is very low relative to the total protein. When immunofluorescence is performed directly with fluorescently labeled antibodies, YOu may need to use higher concentrations of antibodies to achieve similar intensity of indirect immunofluorescence detection (primary antibody plus labeled secondary antibody). In our internal test, it was found that the indirect immunofluorescence staining result was about three times that of the direct immunofluorescence staining signal. Labeled secondary antibodies can still be ligated using Aladdin ® Kit labeled primary antibody, so if multiple antibodies from the same species are used for multicolor immunofluorescence staining, the secondary antibody cannot distinguish between the unlabeled primary antibody from the same species and the use of Aladdin ® Kit labeled primary antibodies. Aladdin ® The labeled antibody can be used as a tertiary staining after detection by traditional indirect immunofluorescence.
