This annealing buffer is not only suitable for the routine annealing of DNA oligos but is especially designed for the annealing of DNA oligos used in RNAi (also known as siRNA) plasmid construction, which are often difficult to anneal. DNA oligos used for RNAi plasmid construction typically contain about 20 complementary sequences are and prone to forming hairpin structures, which can interfere with the correct annealing of the two DNA oligos. Using Aladdin's Annealing Buffer for DNA Oligos (10X) can effectively prevent the formation of hairpin structures in DNA oligos, and the annealed oligos can be directly ligated to purified plasmids that have been digested with restriction enzymes. This typically results in a high yield of positive clones after transformation. The operation with this product is very simple; just mix the DNA oligos to be annealed with the Annealing Buffer for DNA Oligos (10X) in a certain proportion and place the mixture in a PCR machine. The annealing process can be completed in about 20 minutes.
