阿拉丁生产的UltraBio™ Antioxidant (400X)，即UltraBio™抗氧化剂(400X)，是一种使用中性pH值的PAGE凝胶电泳时，用于防止还原性蛋白样品在电泳时被氧化，从而保持电泳过程中蛋白质还原状态的抗氧化剂。本产品适用于中性pH值的预制胶，如阿拉丁的UltraBio™ Plus系列或Bis-Tris系统预制胶的聚丙烯酰胺凝胶的还原性蛋白电泳。在内槽(上槽)电泳液中加入抗氧化剂，可维持样品在电泳过程中的还原状态和条带的最大清晰度。在中性pH值的凝胶中，上样缓冲液中的二硫苏糖醇(Dithiothreitol, DTT)或β-巯基乙醇(β-Mercaptoethanol, β-ME)倾向于留在凝胶顶部，而不能和蛋白一起迁移通过凝胶，这样电泳时部分样品可能会再次被氧化，并导致条带扩散。本抗氧化剂可随蛋白质完全迁移，并在电泳过程中维持蛋白质的还原状态，可弥补DTT等还原剂的不足。4℃保存，一年有效。-20℃可以保存更长时间。在低温保存环境下，本产品可能会有晶体析出，属于正常情况，可室温溶解并混匀后使用。首次使用时可以适当分装，以便于后续使用。UltraBio™ Antioxidant (400X)只适用于中性凝胶系统中，不能用于高pH的凝胶电泳。本产品配制的电泳液只需加入电泳内槽(上槽)，不建议外槽(下槽)也使用，因为会产生较高电流和不必要的浪费。本产品配制的电泳液尽量不回收使用。本产品本身不能有效地还原蛋白中的二硫键，因此不能用于作为还原剂来使用。还原剂推荐使用阿拉丁UltraBio™ Sample Reducing Agent (10X) 。对于非还原性蛋白进行电泳，电泳时不需要使用本产品。不推荐将还原性蛋白样品和非还原性蛋白样品一起进行电泳，如需一起进行电泳，要将两种蛋白加样孔隔开，防止还原性蛋白样品影响非还原性蛋白样品，且电泳液中不可加入本产品。本产品仅限于专业人员的科学研究用，不得用于临床诊断或治疗，不得用于食品或药品，不得存放于普通住宅内。为了您的安全和健康，请穿实验服并戴一次性手套操作。 使用说明 ： 1.在内槽(上槽)电泳液(Cathode buffer)约200ml中性凝胶电泳液如 MES SDS Running Buffer 或 MOPS SDS Running Buffer 中，加入0.5ml Antioxidant (400X)，适当搅拌即可。2.详细的Western操作可以参考阿拉丁相关网页：http://www.aladdin-e.com/support/western.htm。 Aladdin's UltraBio™ Antioxidant (400X) is generally used to prevent reducing proteins from being oxidized during electrophoresis with PAGE gels of neutral pH. This product is suitable for reducing protein electrophoresis with polyacrylamide gels with neutral pH, such as UltraBio™ Plus gel series or Bis-Tris precast gels.The addition of antioxidants to the running buffer in inner chamber can maintain proteins in reduced state to achieve the maximum clarity of protein bands. In gels with neutral pH, Dithiothreitol (DTT) or β-Mercaptoethanol (β-ME) in loading buffer tend to remain on top of the gel rather than migrate with proteins through the gel. Thus, some proteins may be re-oxidized during electrophoresis, which causes band diffusion. However, this antioxidant can migrate with proteins and maintain proteins in reduced state during electrophoresis, making up for the deficiency of reducing agents such as DTT. Precautions ： This product may precipitate at low temperatures, which is normal. The precipitates can be dissolved at room temperature before use. We recommend storing this product in aliquot to avoid repeated freeze-thaw. Antioxidant (400X) is only suitable for neutral electrophoresis system, not for high pH system.Add this product to the running buffer in inner chamber only. Do not add it in the outer chamber as it will produce higher current and unnecessary waste. Running buffer with this product should not be reused.This product cannot effectively reduce disulfide bonds in proteins. We recommended using Sample Reducing Agent (10X) for protein reduction.For electrophoresis of non-reducing proteins, this product is not required.We do not recommend running reducing protein samples and non-reducing protein samples on the same gel. If it is necessary to perform such an experiment, separate reducing samples from non-reducing samples by several loading wells to avoid cross contamination. This product cannot be added into electrophoresis buffer.This product is for R&D only.
