生化机理

ANGPTL3 是血管生成素样蛋白家族的成员，该家族还包括参与调节脂质代谢的 ANGPTL4 和 ANGPTL8。ANGPTL3 编码一种只在肝脏中产生并分泌到血液循环中的蛋白质。在肝脏中，ANGPTL3 只由肝细胞产生，其表达受氧固醇激活的肝 X 受体（LXR）调节。研究表明，可抑制体内和/或体外 ANGPTL3 mRNA 表达的因素包括 PPARδ、他汀类药物、胰岛素、瘦素、甲状腺激素和脂多糖等。ANGPTL3 蛋白结构包含一个介导同源异构化形成的氨基端盘卷结构域（CCD）、一个介导配体活性的羧基端纤维蛋白原样结构域（FLD）以及一个介于 CCD 和 FLD 之间的连接结构域。CCD 前面还有一个高度疏水的区域，这是典型的引导蛋白质分泌的信号肽序列。以往的研究表明，ANGPTL3 的生物效应主要体现在两个方面：调节血管生成和脂质代谢。它对血管生成有微弱的诱导作用，但通过抑制脂蛋白脂肪酶（LPL）的活性，对脂质代谢有显著的调节作用。
