硅谷酸结合型免疫球蛋白样凝集素（Siglec）在介导细胞-细胞相互作用和维持免疫细胞静息状态方面发挥作用。它能优先识别并结合α-2,3-，更热衷于结合α-2,6-连接的含硅酸的聚糖。与 C1q 或乙酰化糖蛋白等配体结合后，CD33 细胞质尾部的两个基于免疫受体酪氨酸的抑制基团（ITIM）会被 LCK 等 Src 类激酶磷酸化。这些磷酸化为蛋白酪氨酸磷酸酶 PTPN6/SHP-1 和 PTPN11/SHP-2 的招募和激活提供了对接位点。反过来，这些磷酸酶又通过信号分子的去磷酸化来调节下游通路。CD33 对单核细胞活化的抑制作用之一需要磷酸肌酸 3- 激酶/PI3K。
