真核生物翻译起始因子 3（eIF-3）复合物的组成成分，蛋白质合成起始的几个步骤都需要它。eIF-3 复合物与 40S 核糖体结合，促进 eIF-1、eIF-1A、eIF-2:GTP:蛋氨酰-tRNAi 和 eIF-5 的招募，形成 43S 预启动复合物（43S PIC）。eIF-3 复合物刺激 mRNA 招募到 43S PIC，并扫描 mRNA 以识别 AUG。终止后核糖体复合物的分解和再循环也需要 eIF-3 复合物，它还能防止 40S 和 60S 核糖体亚基在启动前过早连接。
