核受体辅激活剂，直接与核受体结合，以激素依赖的方式刺激转录活动。参与不同核受体的共激活，如类固醇（PGR、GR 和 ER）、类视黄醇（RXRs）、甲状腺激素（TRs）和前列腺素（PPARs）。还参与 STAT3、STAT5A、STAT5B 和 STAT6 转录因子介导的共激活。对 H3 和 H4 具有组蛋白乙酰转移酶活性；但这种活性的相关性仍不清楚。在创建通过重塑染色质发挥作用的多亚基辅激活剂复合物中发挥核心作用，可能同时参与染色质重塑和一般转录因子的招募。需要与 NCOA2 一起控制白色和棕色脂肪组织之间的能量平衡。需要介导类固醇激素反应。与同工酶 1 和同工酶 3 相比，同工酶 2 具有更高的甲状腺激素依赖性转录活性。
