生化机理

组蛋白去甲基化酶，可使组蛋白 H3 的 "Lys-4"（H3K4me）和 "Lys-9"（H3K9me）去甲基化，从而根据具体情况充当辅助激活剂或核心抑制剂。通过 FAD 氧化底物生成相应的亚胺，然后水解。通过介导 H3K4me（表观遗传转录激活的特异性标记）的去甲基化，发挥核心抑制因子的作用。能使单甲基化（H3K4me1）和双甲基化（H3K4me2）的 H3K4me 去甲基化。可能在抑制神经元基因方面发挥作用。它不能单独使核小体上的 H3K4me 去甲基化，需要 RCOR1/CoREST 的存在才能实现这种活性。它还充当雄激素受体（ANDR）依赖性转录的辅助激活剂，被招募到 ANDR 靶基因上，介导 H3K9me 的去甲基化，H3K9me 是表观遗传转录抑制的一个特定标记。含有 ANDR 的复合物中存在 PRKCB，它会介导组蛋白 H3（H3T6ph）'Thr-6'的磷酸化，而 H3T6ph 是阻止 H3K4me 去甲基化的特异性标记，因此会阻止 KDM1A 的 H3K4me 去甲基化酶活性。对 p53/TP53 的二甲基化 "Lys-370 "进行去甲基化，从而阻止 p53/TP53 与 TP53BP1 的相互作用，抑制 p53/TP53 介导的转录激活。去甲基化并稳定 DNA 甲基化酶 DNMT1。胚胎发生过程中的胃形成所必需。
