组蛋白去甲基化酶能特异性地使组蛋白 H3 的 "Lys-9 "和 "Lys-36 "残基去甲基化，从而在组蛋白编码中发挥核心作用。不对组蛋白 H3 的'Lys-4'、H3 的'Lys-27'和 H4 的'Lys-20'进行去甲基化。能使三甲基化的 H3 "Lys-9 "和 H3 "Lys-36 "残基去甲基化，但对单甲基化和二甲基化残基没有活性。Lys 残基的去甲基化会产生甲醛和琥珀酸。分别通过招募组蛋白去乙酰化酶和 NCOR1 参与 ASCL2 和 E2F 响应启动子的转录抑制。同工酶 2：对肌肉分化至关重要，通过引导去除 Myog 启动子上的抑制性染色质标记，促进 Myog 基因的转录激活。缺少 N 端去甲基化酶结构域。
