生化机理

作为一种 "翻译增强剂"，可将特定的 mRNA 推向多聚体，从而提高蛋白质合成的效率。它与翻译机制和目标 mRNA 的结合可增加每分子 mRNA 的启动次数，并间接稳定 mRNA。与 IGF2 mRNA、MYOD1 mRNA、ARBP/36B4 核糖体蛋白 mRNA 及其自身 mRNA 结合。通过翻译上调 IGF2 的表达，对骨骼肌分化程序至关重要（相似）。通过特异性结合 miRNA 前体 let-7（pre-let-7），抑制 microRNA（miRNA）的生物生成。通过结合 pre-let-7 并招募 ZCCHC11/TUT4 尿苷酸基转移酶，导致 pre-let-7 的末端尿苷酸化。经尿苷酸化的 pre-let-7 miRNA 无法被 Dicer 处理，会发生降解。胚胎干（ES）细胞中pre-let-7的降解有助于ES细胞的维持。与此相反，miR-125 下调神经干细胞中的 LIN28A 可使 pre-let-7 得到处理。特别识别 pre-let-7 末端环路中的 5'-GGAG-3' motif。还能识别并结合末端环中含有 5'-GGAG-3' motif 的非 pre-let-7 pre-miRNA，导致其末端尿苷化并随后降解。
