E3 泛素蛋白连接酶，可将折叠错误的伴侣蛋白底物靶向蛋白酶体降解。与 Hsp70 和 Hsp40 一起泛素化 NOS1。调节几种伴侣复合物的活性，包括 Hsp70、Hsc70 和 Hsp90。介导非规范的短泛素链转移到 HSPA8 上，但对 HSPA8 的降解没有影响。介导 DNA 聚合酶 beta（POLB）在 "Lys-41"、"Lys-61 "和 "Lys-81 "处的多泛素化，从而在碱基切除修复中发挥作用：通过放大 HUWE1/ARF-BP1 依赖性单泛素化催化多泛素化，并导致蛋白酶体降解 POLB。介导 CYP3A4 的多泛素化。
