分解线性和支链多泛素聚合物，明显偏好支链聚合物。参与非锚定'Lys-48'连接的多泛素的分解。以较低的亲和力结合线性和'Lys-63'连接的多泛素。敲除 USP5 会导致 p53/TP53 积累，并增加 p53/TP53 的转录活性，因为积累的非锚定多泛素能够与泛素化的 p53/TP53 竞争，但不能与 MDM2 竞争蛋白酶体识别。
