生化机理

受体酪氨酸激酶与表皮生长因子家族的配体结合，激活多个信号级联，将细胞外线索转化为适当的细胞反应。已知的配体包括 EGF、TGFA/TGF-α、amphiregulin、epigen/EPGN、BTC/betacellulin、epiregulin/EREG 和 HBEGF/肝素结合 EGF。配体结合会引发受体同源和/或异源二聚化，并在关键的细胞质残基上发生自身磷酸化。磷酸化的受体会招募 GRB2 等适配蛋白，进而激活复杂的下游信号级联。激活至少 4 个主要的下游信号级联，包括 RAS-RAF-MEK-ERK、PI3 激酶-AKT、PLCgamma-PKC 和 STATs 模块。还可能激活 NF-kappa-B 信号级联。还可直接磷酸化 RGS16 等其他蛋白，激活其 GTPase 活性，并可能将 EGF 受体信号转导与 G 蛋白偶联受体信号转导结合起来。还能使 MUC1 磷酸化，增加其与 SRC 和 CTNNB1/beta-catenin 的相互作用。同工酶 2 可作为 EGF 作用的拮抗剂。
