电压门控钾通道,介导兴奋膜(主要在大脑中)的跨膜钾转运。有助于调节中枢神经系统神经元的快速动作电位复极化和持续高频发射。同型四聚体通道介导延迟整流电压依赖性钾电流,在高阈值电压下快速激活并缓慢失活。形成四聚体通道,钾离子根据其电化学梯度通过该通道。通道根据膜上的电压差在打开和关闭构象之间交替(可能)。可形成功能性的同源四聚体和异源四聚体通道,其中含有不同比例的 KCNC1,也可能含有其他家族成员;通道特性取决于通道中的α亚基类型。通道特性可通过与辅助亚基(如 KCNE1、KCNE2 或 KCNE3)的结合或一氧化氮(NO)通过 cGMP 和 PKG 介导的信号级联间接调节,从而减缓延迟整流钾通道的激活和失活。有助于视网膜神经节细胞、丘脑皮质和丘脑上核(SCN)神经元以及海马和新皮质中间神经元高频率地发射一连串持续的非常短暂的动作电位。抑制性海马中间神经元的持续最大动作电位点燃频率受组胺 H2 受体激活的负向调节,其调节方式依赖于 cAMP 和蛋白激酶(PKA)磷酸化。通过在神经末梢产生动作电位(AP)再极化,从而减少尖峰诱发的钙离子流入和 GABA 神经递质释放,在维持新皮层 GABA 能中间神经元突触传递的保真度方面发挥作用。需要在新皮层中实现伽马振荡的远距离同步。以光依赖的方式调节嗜铬细胞上核(SCN)神经元自发动作电位发射的昼夜节律。 翻译后:cAMP 依赖性磷酸化抑制通道活性。组胺 H2 受体和 PKA 诱导的磷酸化可延长动作电位尖峰持续时间,降低动作电位尖峰振幅,维持海马中间神经元的最大发射频率;还可降低海马 CA3 锥体细胞层的高频振荡发生率。
