BMS-345541是一种高选择性的IKK-1和IKK-2抑制剂，IC50值分别为4 μM和0.3 μM[1]。 BMS-345541是一种高选择性的IKK抑制剂，在体外和体内均抑制NF-κB依赖的促炎细胞因子的转录。在测量IKK-2催化的GST-IκB的磷酸化实验中，BMS-345541不能抑制其它丝氨酸/苏氨酸和酪氨酸激酶的活性，表明其特异性。这种选择性在细胞中也很明显，BMS-345541只抑制刺激诱导的IκB的磷酸化，而不影响其它信号转导级联反应。由于IKK/ NFκB通路对白血病细胞的活力很重要，因而可用于预测T-ALL的复发。在一些T-ALL细胞系中，BMS-345541可以诱导细胞凋亡和G2/M期细胞的积累。BMS-345541与传统疗法联合使用可用于克服化疗药物的耐受性。 BMS-345541 has been used: as an IκB kinase (IKK) complex inhibitor to study its effects on the interferon-γ (IFN-γ) production by natural killer (NK) cells as an IKK inhibitor to study its effects on the expression of eotaxin and monocyte chemoattractant protein-1 (MCP-1) mRNA in gingival fibroblasts as nuclear factor κB (NF-κB) pathway inhibitor to study its effects on tumor necrosis factor α (TNFα) production in human oral squamous carcinoma cells
