Camostat Mesilate是胰蛋白酶样的蛋白酶抑制剂,抑制上皮细胞钠离子通道(ENaC)功能,IC50为50 nM, 对Trypsin, Prostasin 和 Matriptase作用效果稍弱。A protease and Trypsin inhibitor. Camostat is a trypsin-like protease inhibitor, inhibits airway epithelial sodium channel (ENaC) function with IC50 of 50 nM, less potent to trpsin, prostasin and matriptase. A protease and Trypsin inhibitor.
生化机理
研究发现,甲磺酸卡莫司他是一种蛋白酶抑制剂,能显著提高狗的碳酸氢盐浓度、分泌率和胰液输出量。此外,研究还发现,这种水诱导和碳酸氢盐分泌的增加是通过直接调节狗胰腺的导管细胞实现的。其他研究表明,甲磺酸卡莫司他能抑制大鼠脊髓 Fos 的表达,而 Fos 是神经元激活的标志。此外,研究还表明,这种化合物可以预防或抑制大鼠模型中由胰腺炎诱发的指压异感/过痛。甲磺酸卡莫司他还能防止胰蛋白酶对微球的攻击,这种攻击会导致降解产物 N-苯甲酰基-dl-精氨酸。甲磺酸卡莫司他是KLK、纤溶酶原、凝血酶原和胰蛋白酶的抑制剂。可逆性抑制气道上皮钠通道(ENaC)功能(IC 50 = 50 nM)。抑制单核细胞产生 TNF-α 和单核细胞趋化蛋白-1 (MCP-1),并干扰胰腺支架的增殖。
