胰凝乳蛋白酶是来源于牛胰的一种丝氨酸蛋白酶，可特异性酶切Tyr、Phe、Trp和Leu的C-末端的肽键。也有少量的Met、Ala、Asp和Glu的C-末端的肽键裂解。其中的残留胰蛋白酶用TLCK灭活后纯化处理。该蛋白酶用Ca²⁺离子激活并稳定 pH 值范围：在PH=7.8时，α-Chymotrypsin蛋白酶具有最佳活性。适用范围：α-Chymotrypsin蛋白酶特异性水解变性后蛋白质Y、F、W和L位。溶液酶解SOP：胰凝乳蛋白酶按照1:50蛋白酶和蛋白的量（w/w）使用，在30℃下于含10mM CaCl₂，pH7.8的100mM Tris-HCI中进行酶切消化2-12小时。注意事项： 1.水浴温度高于37°C时蛋白酶会自我水解。 2.样品消化过程中尿素和盐酸胍的浓度最好小于1M。
