生化机理

组织溶梭菌产生两类胶原酶。梭菌胶原酶比哺乳动物胶原酶更高效。它能够在胶原蛋白三螺旋中的多个切割位点进行切割。它与气性坏疽中的细菌入侵有关。从组织中有效释放细胞需要胶原酶和中性蛋白酶的作用。每摩尔酶可被4克原子钙（Ca 2+ ）活化。培养物滤液被认为含有至少7种不同蛋白酶，分子量范围为68-130kDa。最适pH为6.3-8.8。该酶通常用于消化组织样品中的结缔组分，以释放单个细胞。胶原酶处理可导致一些细胞死亡。通常使用0.1-5 mg/mL的浓度进行消化。反应持续时间从15分钟到几小时不等，以产生令人满意的细胞解离效率，同时又不会引起太多的细胞死亡。含有钙和BSA的Krebs Ringer缓冲液是首选，并且需要添加Zn 2+ 以确保活性。在测试中，对于每个大鼠肝脏，使用该酶以约1 mg/mL的浓度、总体积100mL释放肝细胞。每摩尔胶原蛋白酶被4g原子钙活化。它被乙二醇-双（β-氨基乙基醚）-N，NN′,N′-四乙酸，β-巯基乙醇、谷胱甘肽、巯基乙酸和8-羟基喹啉抑制。
