软骨素酶B(Chondroitinase B)来源于肝素黄杆菌（Flavobacterium heparinum），可降解硫酸皮肤素。此酶通过β消除机理作用于二糖重复单元之间的β1-4糖苷上并将其切断。降解反应的产物为寡糖，主要是不饱和二糖。用途酶解 HPLC 法测定硫酸皮肤含量和二糖组成；肝素、硫酸乙酰肝素的精制；低分子硫酸皮肤素酶法制备。推荐使用条件最适pH：8.0 pH适用范围：5.5-9.5 最适温度：37 ℃ 温度适用范围：20~50 ℃ 使用缓冲液：20mmol/L Tris-HCl (pH:8.0),150 mmol/L NaCl, 5mmol/L CaCl2

生化机理

Cleaves the glycosaminoglycan, dermatan sulfate. Catalytic activity: Eliminative cleavage of dermatan sulfate containing (1->4)-beta-D-hexosaminyl and (1->3)-beta-D-glucurosonyl or (1->3)-alpha-L-iduronosyl linkages to disaccharides containing 4-deoxy-beta-D-gluc-4-enuronosyl groups to yield a 4,5-unsaturated dermatan-sulfate disaccharide (DeltaUA-GalNAc-4S).

生物活性

≥200 U/mg protein；单位定义：一个酶活性单位是指以硫酸软骨素B为底物，在 pH 8.0、温度 30℃ 条件下，每小时形成 0.1μmol 不饱和糖醛酸所需的酶量。
