硫酸软骨素酶AC (Chondroitinase AC) 来源于肝素黄杆菌（Flavobacterium heparinum），可降解硫酸软骨素A、硫酸软骨素C。此酶通过β消除机理作用于二糖重复单元之间的β1-4糖苷上并将其切断。降解反应的产物为寡糖，主要是不饱和二糖。用途：酶解 HPLC法测定硫酸软骨素含量和二糖组成；低分子硫酸软骨素酶法制备；肝素、硫酸乙酰肝素的精制。推荐使用条件：最适pH：8.0 pH适用范围：5.5-9.5 最适温度：37 ℃ 温度适用范围：10~70 ℃ 使用缓冲液：20mmol/L Tris-HCl (pH:8.0),150 mmol/L NaCl, 5mmol/L MgCl2

生化机理

Catalytic activity: Eliminative degradation of polysaccharides containing 1,4-beta-D-hexosaminyl and 1,3-beta-D-glucuronosyl linkages to disaccharides containing 4-deoxy-beta-D-gluc-4-enuronosyl groups. Cofactor: Ca2+. Note: Binds 1 Ca2+ ion per subunit.
