钙离子浓度测量对众多生物学研究至关重要。水母素（aequorin）复合体由22 kDa的脱辅基水母素蛋白、分子氧以及发光基团腔肠素组成。当三个Ca²⁺离子与该复合体结合时，腔肠素被氧化为腔肠酰胺，同时释放二氧化碳和蓝光。水母素生物发光强度与Ca²⁺浓度约呈三次方依赖关系，使其检测范围可覆盖~0.1 µM至>100 µM的宽浓度区间。与荧光钙离子指示剂不同，Ca²⁺结合的水母素无需样品激发光即可检测，从而避免了自发荧光的干扰。天然腔肠素及其衍生物会赋予水母素复合体不同的Ca²⁺亲和力与光谱特性。研究表明，采用腔肠素hcp重组脱辅基水母素能获得最佳综合发光性能，兼具高量子产率和快速响应时间。但腔肠素类似物在胞内重组水母素的过程可能较缓慢。相较于天然腔肠素，使用cp、f或h型腔肠素重构的水母素发光强度可提升10-20倍。其中，腔肠素h和cp已被用于G蛋白偶联受体（GPCR）的高通量筛选实验。适用范围用于钙离子检测产品优势 1.腔肠素（Coelenterazine）在与钙结合后被氧化为腔肠酰胺（Coelenteramide），同时释放出二氧化碳并发出‘蓝色’光。 2.化学发光/生物发光，无需激发光源，背景噪声低，灵敏度高。 3.腔肠素属于瞬时发光，发光过程极为迅速，持续时间在微秒级别。

生化机理

腔肠素 hcp 的发光强度是原生腔肠素的 190 倍，反应时间也比原生腔肠素快。
