CS/DS硫酸酯酶是一类可以专一性水解去除CS/DS糖链中特定部位硫酸基团的酶类，是研究CS/DS构效关系的重要工具酶，包括GalNAc-4- O -硫酸酯酶、6- O -硫酸酯酶和GlcUA/IdoUA-2- O -硫酸酯酶等。与糖胺聚糖降解酶类似，糖胺聚糖硫酸酯酶也可以分为两类：内切型糖胺聚糖硫酸酯酶和外切型糖胺聚糖硫酸酯酶。外切型硫酸酯酶只对CS/DS糖链末端的硫酸根有脱去作用，作用于CS/DS经降解酶降解产生的二糖以及低分子量寡糖，主要参与CS/DS的代谢过程；而内切型硫酸酯酶不仅可以作用于糖胺聚糖末端的硫酸根，对内部的硫酸根也有脱去作用，因此可以有效地的作用于多糖，从而对多糖的内部结构进行修饰调节。
