我们利用硫酸软骨素酶选择性降解各种硫酸软骨素/硫酸皮肤素，并结合凝胶过滤和阴离子交换HPLC，制备了具有各种硫酸化模式的高纯C4，5不饱和二糖标准。包括硫酸软骨素/硫酸皮肤素经软骨素酶降解后常见七种不饱和二糖：Δdi-0S （ΔUA-[1→3]-GalNAc），Δdi-4S（ΔUA-[1→3]-GalNAc-4S），Δdi-6S（ΔUA-[1→3]-GalNAc-6S），Δdi-4S6S（ΔUA-[1→3]-GalNAc-4S-6S），Δdi-2S4S（ΔUA-2S-[1→3]-GalNAc-4S），Δdi-2S6S（ΔUA-2S-[1→3]-GalNAc-6S）和Δdi-2S4S6S（ΔUA-2S-[1→3]-GalNAc-4S-6S及其相应的2-Aminobenzamide （2AB）荧光标记二糖标准. We used chondroitinase to selectively degrade various chondroitin sulfate/dermatan sulfate, combined with gel filtration and anion exchange HPLC, to prepare high-purity C4, 5 unsaturated disaccharides standards with various sulfation modes. Including seven common unsaturated disaccharides after chondroitin sulfate/dermatan sulfate degradation by chondroitinase: Δ di-0S (Δ UA - [1 → 3] - GalNAc), Δ di-4S (Δ UA - [1 → 3] - GalNAc-4S), Δ di-6S (Δ UA - [1 → 3] - GalNAc-6S), Δ di-4S6S (Δ UA - [1 → 3] - GalNAc-4S), Δ di-2S4S (Δ UA-2S - [1 → 3] - GalNAc-6S), and Δ di-2S4S6S (Δ UA-2S - [1 → 3] - GalNAc-6S) 3] - GalNAc-4S-6S and its corresponding 2-Aminobenzamide (2AB) fluorescently labeled disaccharide standard
