内切-β-半乳糖苷酶，E.C.：3.2.1.103，来源于 Bacteroides fragilis，内切β-半乳糖苷酶在大肠杆菌中重组表达的，是一种水解聚 n -乙酰乳胺结构中低聚糖（LNnT）内部β-半乳糖苷键的酶。对牛角膜硫酸角蛋白、牛奶低聚糖和糖脂类乳酸-n -新四糖基神经酰胺和乳酸-n -四糖基神经酰胺具有特异性。使用方法 1.在反应体系中依次加入底物，50 mM 醋酸钠, pH 5.8，加入 5 mM CaCl2 混匀，加入适量内切-β-半乳糖苷酶。用去离子水定容到反应体积。 2.将样品置于 37℃孵育 2-4 小时或者过夜。【注】体系放大时候，孵育时间或酶量需要根据实际情况调整，可低速震荡混匀，可以增加转化率。注意事项为了您的安全和健康，请穿实验服并戴一次性手套操作。本产品仅作科研用途！

生化机理

未分枝的重复聚乙酰乳糖胺中的内部β(1-4)半乳糖连接[GlcNAc β(1-3)Gal β (1-4)] 结构是优选的底物。诸如硫酸角质素的硫酸化结构也会被裂解。底物的支化或岩藻糖基化可减少或完全抑制裂解。半乳糖上C-6的硫酸化将阻断裂解。取决于与优选底物的偏差，新乳酸基团的低聚糖会以极低的速率而被裂解。例如，Gal β(1-3)GlcNAc β(1-3) Gal β(1-4)Glc会以硫酸角质素5X10的速率而被裂解β-半乳糖苷酶可将乳糖切割成单糖成分、葡萄糖和半乳糖。它还在反馈环中催化葡萄糖转糖基化为β-半乳糖苷酶的诱导物异乳糖。

生物活性

≥1000 U/mg protein，单位定义：在37℃, pH 5.8 条件下，每小时从LNnT 中释放 1nmol还原糖的酶量。
