产品介绍

固定剂、破膜剂用于细胞内细胞因子染色前对细胞膜的处理。固定剂起稳定细胞膜、保持细胞膜表面抗体与抗原结合的作用；破膜剂使流动的、完整的细胞膜产生小孔以利于抗体进入细胞。用流式细胞仪检测细胞内抗原及DNA时，对待测细胞进行固定、破膜处理后，加入相应抗体或检测试剂，孵育一定时间即可上机分析。此核膜破膜剂是10倍浓缩液，使用时需根据具体的实验要求用PBS稀释后使用。使用说明： 1、试剂准备：取1ml破膜剂，加入到9ml PBS中，备用。 2、至少收集1×10⁶ 待测细胞，1500rpm离心5min，弃上清。 3、加入500μl固定剂。 4、固定10min。 5、1500 rpm 离心5min。 6、弃上清，加入PBS洗一次，1500rpm离心5min。 7、弃上清，加入1ml破膜剂。 8、室温孵育10-15min。 9、1500 rpm 离心5min。 10、弃上清，加入100μl破膜液重悬细胞。 11、加入胞核检测抗体进行染色后，PBS洗涤后，用流式细胞仪进行分析。注意事项： 1、如果有膜蛋白，请在步骤3前，先染膜蛋白，再做步骤3的固定 2、在第10步骤，可以用染色缓冲液重悬细胞，但是我们建议用破膜液稀释抗体，效果会更好。
