大肠杆菌产生的β-葡萄糖醛酸酶用于尿液和其他液体中β-葡萄糖醛酸的酶促水解。它不会水解α-葡萄糖醛酸或β-葡萄糖苷。该酶来自大肠杆菌。大肠杆菌具有较高的水解活性，并且在水解过程中比对β-葡萄糖醛酸结合物浓度变化更为敏感的类似酶更能保持这种活性。 β-葡萄糖醛酸酶催化以下反应：处理说明在75mM磷酸盐缓冲液中复溶至5mg/ml时，pH6.8，得到澄清至微浑浊的溶液。无论浑浊程度如何，酶均具有活性，可用于代谢物水解。在75mM磷酸盐缓冲液，pH6.8的溶液中，(≥5mg/ml)可在-20℃下储存至2个月，活动性很少或没有丧失。产品应用 1、可用于检测尿液中雄甾酮、17-羟基皮质类固醇和雌三醇的存在。 2、确定α-羟基三唑仑的酶促水解的最佳条件。注：在pH6.8、37°C条件下，每小时可从酚酞葡萄糖苷中释放1.0μg酚酞(30分钟试验）。与天然含有几乎等量β-葡萄糖醛酸酶和硫酸酯酶活性的蜗牛（Helixpomatia）酶制剂不同,大肠杆菌β-葡萄糖醛酸酶制剂基本上不含硫酸酯酶活性。

生化机理

Displays beta-glucuronidase activity with the artificial substrate p-nitrophenyl-beta-D-glucuronide (PNPG) and with 4-methylumbelliferyl-glucuronide. Is likely capable of scavenging glucuronate from a range of chemically distinct xenobiotic and endobiotic glucuronides present in the gastrointestinal (GI) tract, to be able to utilize these diverse sources of carbon. As part of the GI microbiome, this enzyme is able to reactivate glucuronide drug conjugates, such reactivated compounds can significantly damage the GI tract.
