产品介绍

单克隆抗体是通过杂交瘤技术进行生产，即非分泌性骨髓瘤细胞系与产生抗体的B淋巴细胞融合。融合后，存活的杂交瘤细胞比例较低。因此，需要以牺牲亲代细胞为代价，采用选择性培养基（如HAT培养基）来促进杂交瘤细胞的存活。 HAT培养基适用于针对未融合或自融合的HGPRT–骨髓瘤细胞进行融合后选择。次黄嘌呤和胸苷提供预先形成的嘌呤和嘧啶，用于杂交瘤通过利用脾细胞融合形成的HGPRT的补救途径进行DNA合成。叶酸拮抗剂氨基蝶呤可从头抑制核苷生物合成途径。本品含有：次黄嘌呤(5mM)、氨基蝶呤(20µM)和胸腺嘧啶核苷(0.8mM)。10mL本品可制备500mL培养基。各成分最终工作浓度为：100µM次黄嘌呤、0.4µM氨基蝶呤、16µM胸腺嘧啶核苷。使用说明：根据实验需求取适量本品，按照50倍稀释添加到细胞培养基中。即1mL本品可配制50mL细胞培养基。注意事项： 1、本产品为50×浓缩液，请根据需要稀释后使用。 2、若本品在室温解冻后有沉淀析出，可在37℃水浴中重新溶解，摇匀后使用。 3、本产品为无菌包装，请注意无菌取用。 4、为了您的安全和健康，操作时请穿着实验服并佩戴一次性手套。
