KPT-276是KPT-185的类似物,是一种CRM1和核外运选择性抑制剂(SINE). 染色体维持蛋白1(CRM1)是一种核外运受体,参与转录因子\细胞周期调控子\癌症抑制子和RNA分子的主动转运.在癌症中,CRM1过表达并过度转运. KPT-276是一种选择性CRM1抑制剂,不可逆地结合CRM1,阻断CRM1介导的核外运.在人类多发性骨髓瘤(MM)细胞系(HMCLs)中,KPT-276可不可逆地和特异性地抑制编码CRM1的XPO1核外运,并显著减少HMCLs的活力.在从MM病人分离的骨髓细胞中,KPT-276可诱发细胞凋亡.此外,KPT-276可下调c-MYC\CDC25A和BRD4的表达,引起G1/S期停滞. 在人类急性髓性白血病(AML)异种移植物小鼠模型中,KPT-276可显著降低小鼠存活率,并减少白细胞数量.另外,KPT-276可显著降低脾脏重量和FLT3蛋白表达[1].在异种移植MM小鼠模型中,KPT-276可抑制肿瘤生长.
