N-hydroxylsuccinimide (NHS) functionalized polyethylene glycol (PEG-NHS) is an amino (-NH 2 ) reactive PEG derivative that can be used to modify protein, peptide or any other surfaces with their available amino groups. NHS esters react with primary amine groups at pH 7~8.5 to form stable amide bonds.Compared to other PEG NHS ester derivatives, our succinimidyl carbonate (SC) functionalized mPEG-NHS offers superior reactivity and higher stability in aqueous solution. NHS esters react with deprotonated primary amines, therefore, the reaction requires neutral to basic pH values to proceed. Primary amines react with NHS esters by nucleophilic attack and NHS is released as a byproduct. Hydrolysis of the NHS-ester competes with the reaction in aqueous solution and increases with increasing pH. 生化机理
一般情况下,NHS PEG 摩尔过量 10 至 50 倍于含胺材料的量就能充分缀合。 所需材料 连接缓冲液:pH 为 8.5 的 100 mM 碳酸氢钠缓冲液或 pH 为 7-8.5 的其他无胺缓冲液。 PEG NHS 储存液:100 毫克加入 1 毫升共轭缓冲液中。 洗涤液:蒸馏水或任何水性缓冲液。 反应步骤: 将目标材料溶解在共轭缓冲液中。估计目标材料上伯胺基团的浓度。向目标共轭材料中加入 NHS PEG 原液,最终浓度至少为 10 mg/mL。最佳共轭需要 10~50 摩尔过量的 PEG NHS;将混合物在室温下搅拌 30~60 分钟或在 4 0C 下搅拌 2 小时。共轭物可通过尺寸排阻色谱法或透析法纯化。
